| Description | Purity ~95% (HPLC). A cell-permeable 37-mer peptide derived from the NEMO NOA ubiquitin binding site (294-314 of mouse sequence with D304R), and fused with an Antennapedia cell penetrating protein (A) at the N-terminus. It is shown to attenuate NF-kB activation via blocking interactions between K63-polyubiquitin chains and the NOA ubiquitin binding domain of NEMO (=17uM for CC2-LZ/A-UBI complex). The biotin-labled A-UBI specifically inhibits the LPS-induced NF-kB activation (IC50=1.1uM, compared with 23uM for IKKb derived A-NBD) and TNF-a induced NF-kB activation (IC50=21uM) in 70Z/3-C3 pre-B lymphocytes. It displays an inhibitory effect on nuclear translocation of NF-kB p65 and IKK kinase activity at 10uM, but does not significantly affect other signaling pathways such as MAPK/ERK, MAPK/JNK, NF-kB and NFAT.
