| Description | Epidermal growth factor (EGF) receptor is a 170kD single-pass transmembrane tyrosine kinase. Ligand binding to this receptor results in receptor dimerization, autophosphorylation (in trans), activation of various downstream signaling molecules and lysosomal degradation (1,2). Tyrosine 845 resides in the activation loop of the receptor kinase domain (3,4). Phosphorylation of Tyr845 may stabilize the activation loop, maintain the enzyme in an active state and provide a binding surface for substrate proteins. It has been shown that c-Src is involved in phosphorylation of the EGF receptor on Tyr845, which is associated with modulation of receptor function (5). Phospho-tyrosine 992 of activated EGF receptor is a direct binding site for the phospholipase C-gamma (PLC-gamma) SH2 domain (6).
