| Description | Tetanolysin is one of a family of cholesterol binding toxins, known as cytolysins. These proteins form pores in the cytoplasmic membrane that allow ions and other molecules to pass through. Tetanolysin is purified from Clostridium tetani, and has a molecular weight of ~55kD. Cytolysins, including tetanolysin, generally have an LD50 of less than a microgram for mice, rabbits, and other laboratory animals. Their potent lethal and lytic properties are irreversibly inactivated by cholesterol and inhibited by oxidation. Conversely, the activity is somewhat enhanced by the addition of a reducing agent such as dithiothreitol. Tetanolysin, purified, is hemolytic to rabbit red blood cells at maximum dilutions of 10-5-10-6mg/ml. Tetanolysin is used to permeabilize cellular membranes. Thus, molecules that could not other- wise penetrate a membrane could be introduced to the endoplasm for the study of various cell functions.
